- Research Interests * Isolation, cloning of heparan sulfate proteoglycans and studies of their functions by tissue specific gene knock out in mice. * Structure/function relationship of lipolytic enzymes. * Identification of major enhancers and silencers in lipoprotein lipase and hepatic lipase gene promoters. * Function of lipolytic enzymes and proteoglycans in lipoprotein receptor mediated endocytosis * Function of GPI anchored HDL-binding protein (GPI-HBP-1) in the hydrolysis of triglycerides-rich lipoproteins My laboratory is concerned with various aspects of the biochemistry of lipolytic enzymes and their functions in the genesis of circulating plasma lipoproteins. During the recent past, the laboratory has purified to homogeneity, lipoprotein lipase (LPL) and hepatic lipase (HL) in tissues of human and experimental animals. Panels of monoclonal antibodies against these two lipases have been isolated and cDNA's and genomic sequences for several lipases have been isolated. A major thrust of our efforts is to conduct structural and functional studies of these lipases. Fine mapping of HL and LPL is being conducted by assigning monoclonal antibodies to specific epitopes on proteolytic, cyanogen bromide, chemically generated fragments and to peptides generated in vitro by expression vectors. Characterized monoclonal antibodies are being evaluated for their ability to inhibit functional properties of lipases: Binding of enzyme to lipid interfaces and heparan sulfate; catalytic activity; binding of the activator apoCII to LPL and dimer formation from monomeric LPL. In identifying structural domains, where appropriate, chemical modifications, site directed mutagenesis, and reactivity of monoclonal antibodies to specific fusion products generated in vitro by expression vectors are employed. My laboratory has recently (2006) collaborated with the laboratory of Dr Stephen Young at UCLA in identifying the functions of GPI-HBP-1 (GPI anchored HDL-binding protein). The protein was first identified as an HDL-binding protein. We have discovered that it also binds LPL (lipoprotein lipase), VLDL, HDL and chylomicrons. Our current hypothesis is that GPI-HBP-1 functions as a “lipolysis platform” assembling a complex that would include LPL, substrate (VLDL, chylomicrons) and HDL. During lipolysis, the proximity of HDL would favor the channeling of surface remnants (phospholipids, cholesterol, ApoCs ) from VLDL to HDL. In support of this hypothesis GPI-HBP-1 deficient adult mice and humans are grossly hypertriglyceredemic.
